Overview

This drug is a combination therapy consisting of all-trans-retinoic acid (ATRA) and interferon alpha-2a (IFN-alpha2a). ATRA is a derivative of vitamin A that induces differentiation and inhibits proliferation in certain cancer cells. IFN-alpha2a is a recombinant cytokine with antiproliferative, immunostimulatory, antiviral, and differentiation-potentiating effects. The rationale for combining these agents lies in their complementary mechanisms—ATRA promotes cellular differentiation while IFN-alpha2a enhances immune response and may potentiate the effects of retinoids. This combination has been investigated primarily in pediatric oncology for refractory neuroblastoma and Wilms tumor but did not demonstrate significant clinical activity in phase 1/2 trials[1][2][5]. The regimen was generally well tolerated; headache was the most common toxicity.
