This is a combination therapy consisting of all-trans retinoic acid (ATRA, also known as tretinoin) and prednisone. All-trans retinoic acid is a derivative of vitamin A that acts primarily as an agonist at the retinoic acid receptor alpha (RARα), promoting differentiation and apoptosis in certain malignant cells, most notably in acute promyelocytic leukemia (APL). Prednisone is a synthetic corticosteroid prodrug that is converted to prednisolone in the body; it acts as an agonist at the glucocorticoid receptor, exerting potent anti-inflammatory and immunosuppressive effects. The combination has been studied for its ability to induce remission in APL while reducing pulmonary toxicity associated with ATRA by using corticosteroid prophylaxis[1][2]. It has also been investigated preclinically for potential steroid-sparing effects in autoimmune diseases such as lupus nephritis[2].
