Overview

A combination chemotherapy regimen consisting of all-trans retinoic acid (ATRA), vincristine, dactinomycin, and cyclophosphamide. This regimen is being evaluated by Fudan University for the treatment of intermediate-to-high-risk rhabdomyosarcoma. ATRA, a derivative of vitamin A, acts as a differentiating agent by binding to retinoic acid receptors, potentially inducing the maturation of malignant cells. Vincristine is a vinca alkaloid that inhibits microtubule polymerization, disrupting mitotic spindle formation. Dactinomycin is a cytotoxic antibiotic that intercalates into DNA and inhibits RNA synthesis. Cyclophosphamide is an alkylating agent that forms DNA cross-links, leading to cell death. The combination aims to leverage the differentiation-inducing properties of ATRA alongside the cytotoxic effects of the standard VAC regimen to improve clinical outcomes in patients with rhabdomyosarcoma.
