Overview

Allergen extract from lolium perenne pollen, glutaraldehyde-modified is a specific allergoid used in allergen-specific immunotherapy (AIT). Developed by Inmunotek, it is primarily indicated for the treatment of allergic rhinitis, allergic rhinoconjunctivitis, and allergic asthma triggered by perennial ryegrass (*Lolium perenne*) pollen. The glutaraldehyde modification process cross-links the allergen proteins to form high-molecular-weight polymers known as allergoids. This chemical modification significantly reduces the IgE-binding capacity of the allergens, thereby decreasing the risk of immediate allergic reactions and anaphylaxis during administration. Despite the reduced allergenicity, the T-cell epitopes remain preserved, allowing the drug to effectively induce immune tolerance. The mechanism of action involves shifting the immune response from a Th2-dominated profile to a Th1 and regulatory T-cell (Treg) response, characterized by the production of IL-10 and TGF-beta, and an increase in allergen-specific IgG4 blocking antibodies. It is typically administered via subcutaneous injection and is often adsorbed onto aluminum hydroxide to enhance its immunogenic effect.
