Allergoid-mannan refers to a new generation of hypoallergenic vaccines for allergen immunotherapy. These are polymerized allergen extracts (allergoids) chemically conjugated to nonoxidized mannan, a polysaccharide derived from yeast such as *Saccharomyces cerevisiae*. The conjugation targets dendritic cells by exploiting the interaction between mannan and C-type lectin receptors on these immune cells, leading to rapid uptake and induction of tolerogenic dendritic cells. This process promotes regulatory T cell (Treg) responses and increases IL-10 production, shifting the immune response away from allergy-associated Th2 pathways toward tolerance. Allergoid-mannan vaccines have been developed for various allergens including grass pollen, birch pollen, and house dust mite. Clinical studies show they are safe and effective in reducing allergic symptoms in conditions like allergic rhinoconjunctivitis. The technology was developed by Inmunotek.
