ALLO-319 is an investigational allogeneic chimeric antigen receptor (CAR) T cell therapy being developed by Allogene Therapeutics in collaboration with Overland Pharmaceuticals. It is engineered to target CD75, a carbohydrate antigen that is frequently overexpressed on the surface of various solid tumor cells. As an "off-the-shelf" (allogeneic) therapy, ALLO-319 utilizes T cells derived from healthy donors rather than the patients themselves. These cells are genetically modified to express a CAR specific to CD75 and are typically edited using TALEN technology to disrupt the T-cell receptor (TCR) alpha constant (TRAC) gene to reduce the risk of graft-versus-host disease (GvHD). The therapy is currently being evaluated in Phase 1 clinical trials for the treatment of various solid tumors.
