Overview

ALLO-605 is an allogeneic, gene-edited chimeric antigen receptor (CAR) T cell therapy developed for the treatment of relapsed or refractory multiple myeloma and other B cell maturation antigen (BCMA)-positive malignancies. It is derived from healthy donor T cells and engineered using TALEN technology to express a next-generation anti–BCMA CAR as part of the proprietary TurboCAR platform, which incorporates programmable cytokine signaling to enhance potency and overcome challenges such as T cell exhaustion. Preclinical studies have shown that ALLO‑605 demonstrates enhanced cytokine secretion, polyfunctionality, improved serial killing activity in vitro, and superior antitumor activity compared to standard BCMA CAR-T cells. The therapy is administered following a lymphodepletion regimen including fludarabine, cyclophosphamide, and the anti-CD52 monoclonal antibody ALLO‑647[5][6][7][9].
