ALLO-647 is a humanized monoclonal antibody that targets CD52, a cell surface protein expressed on lymphocytes. By binding to CD52, ALLO-647 induces the lysis of CD52-positive lymphocytes through immune-mediated mechanisms, resulting in immunodepletion. This selective depletion of host lymphocytes is used as part of the conditioning regimen for allogeneic CAR T-cell therapies to prevent graft-versus-host disease (GvHD) and enable engraftment and expansion of donor-derived CAR T cells. The drug has been primarily developed for use in patients with relapsed or refractory large B-cell lymphoma (LBCL) and follicular lymphoma as part of an allogeneic CAR T-cell therapy protocol[1][3][4][5][6].
