ALLO-715 + nirogacestat is an investigational combination therapy consisting of ALLO-715, an allogeneic anti–B-cell maturation antigen (BCMA) chimeric antigen receptor (CAR) T cell therapy, and nirogacestat, a small-molecule gamma-secretase inhibitor. ALLO-715 is engineered using TALEN gene-editing to delete TRAC and CD52 genes, reducing risk of graft-versus-host disease and enhancing lymphodepletion. Nirogacestat increases cell-surface BCMA expression, potentially enhancing anti-myeloma efficacy of BCMA-targeting therapies. The combination is being studied for relapsed/refractory multiple myeloma. ALLO-715 is developed by Allogene Therapeutics; nirogacestat is developed and manufactured by SpringWorks Therapeutics[1][2][4][5][6].
