Allo15MCAR-NKT is an allogeneic, off-the-shelf cellular immunotherapy consisting of invariant natural killer T (iNKT) cells engineered to express a chimeric antigen receptor (CAR) targeting mesothelin (MSLN) and a secreted form of interleukin-15 (IL-15). The therapy is produced using a proprietary platform that involves lentiviral engineering of human hematopoietic stem and progenitor cells (HSPCs) followed by a multi-stage ex vivo differentiation process. Allo15MCAR-NKT cells utilize a multimodal anti-tumor mechanism: the CAR (utilizing the SS1 scFv) targets MSLN-expressing tumor cells; the invariant TCR recognizes the non-polymorphic CD1d molecule on immunosuppressive cells such as tumor-associated macrophages (TAMs) and myeloid-derived suppressor cells (MDSCs); and intrinsic natural killer receptors (e.g., NKG2D, DNAM-1) provide additional cytotoxicity. The integrated IL-15 sequence promotes the expansion, persistence, and metabolic fitness of the cells within the tumor microenvironment. Preclinical data indicates high efficacy in uterine endometrial carcinoma and ovarian cancer models with a favorable safety profile, showing no evidence of graft-versus-host disease (GvHD) and minimal risk of cytokine release syndrome (CRS).
