AlloCAR-NKT is an investigational allogeneic cellular therapy comprised of natural killer T cells (NKT) genetically engineered to express a chimeric antigen receptor (CAR), allowing them to target cancer cells via several mechanisms[1]. These cells can recognize and kill tumor cells through a combination of CAR recognition (of a specific tumor antigen), T-cell receptor (TCR) engagement (primarily recognizing CD1d), and natural killer receptor (NKR) activation (binding to NK ligands such as NKG2D and DNAM-1 ligands on tumor cells), enabling triple- or double-targeting of both hematologic malignancies and solid tumors. AlloCAR-NKT is designed for off-the-shelf use, generated from healthy donor hematopoietic stem cells, and may overcome antigen escape and reduce cytokine release syndrome compared to conventional CAR-T therapies[1].
