AlloCAR70-NKT is an allogeneic, "off-the-shelf" chimeric antigen receptor (CAR)-engineered invariant natural killer T (iNKT) cell therapy. It is derived from cord blood hematopoietic stem and progenitor cells (HSPCs) and engineered to target CD70, a protein often overexpressed in acute myeloid leukemia (AML) and other myeloid malignancies. The therapy utilizes the unique properties of iNKT cells, which possess an endogenous T-cell receptor (TCR) that recognizes CD1d, providing a dual-antigen recognition system when combined with the CD70-specific CAR. Preclinical research indicates that treatment with hypomethylating agents (HMAs) can sensitize AML cells to AlloCAR70-NKT by upregulating CD70 and CD1d. This product is designed to avoid graft-versus-host disease (GvHD) and provide sustained anti-tumor activity with improved persistence compared to traditional peripheral blood-derived NKT cells.
