Allocetra-OTS is an off-the-shelf, allogeneic cell-based immunotherapy consisting of mononuclear cells (PBMCs) from healthy donors induced into an early apoptotic state. Developed by Enlivex Therapeutics, the therapy is designed to reprogram macrophages and restore immune homeostasis by leveraging the natural 'eat me' signal, phosphatidylserine (PtdSer), expressed on the surface of the apoptotic cells. When these cells are engulfed by host macrophages and other antigen-presenting cells, they trigger a shift from a pro-inflammatory or immunosuppressive state toward a homeostatic or anti-tumor phenotype. In oncology, Allocetra-OTS is being investigated for its ability to modulate the tumor microenvironment, particularly in peritoneal cancers and other solid tumors, often in combination with immune checkpoint inhibitors like anti-PD-1 antibodies.
