Drug intelligence / Profile preview
AlloCol is a proprietary oral small molecule formulation of cyclosporine developed by Sigmoid Pharma (now Sublimity Therapeutics) for the prevention and treatment of Graft versus Host Disease (GvHD). The product utilizes the SmPill drug delivery platform to deliver solubilized cyclosporine in a dual immediate- and controlled-release format, designed to optimize the drug's pharmacokinetic profile and local/systemic distribution. Cyclosporine, the active pharmaceutical ingredient, is an immunosuppressant that functions by binding to cyclophilin and subsequently inhibiting the phosphatase activity of calcineurin. This inhibition prevents the dephosphorylation of the nuclear factor of activated T-cells (NFAT), thereby blocking the transcription of inflammatory cytokines such as interleukin-2 and suppressing T-cell activation. AlloCol has been granted Orphan Drug Designation by the FDA and has successfully completed Phase 1 pharmacokinetic clinical trials.
