Allogeneic Adoptive Immune Therapy (AAIT) is a specialized cellular therapy developed by researchers at the Fifth Medical Center of PLA General Hospital (formerly Beijing 302 Hospital) in Beijing, China. The therapy is designed for patients with advanced acquired immunodeficiency syndrome (AIDS), particularly those with severe immunosuppression (CD4+ T-cell counts < 200 cells/μl) who remain at high risk for opportunistic infections despite antiretroviral therapy (ART). The procedure involves mobilizing peripheral blood mononuclear cells (MNCs) in healthy, HLA-mismatched donors using recombinant granulocyte-colony-stimulating factor (G-CSF). These mobilized cells (G-MNCs) are then collected via leukapheresis and transfused into the patient. The mechanism of action involves the rapid restoration of cellular immunity through the introduction of functional donor immune cells, which helps control opportunistic infections and potentially stimulates the patient's own immune reconstitution. Clinical evaluation has progressed through Phase 1 and Phase 2a trials, demonstrating preliminary safety and efficacy in improving survival and immune markers in advanced AIDS patients.
