This investigational dendritic cell-based vaccine, developed by Roswell Park Cancer Institute, is designed for the treatment of recurrent prostate cancer. The therapy utilizes autologous dendritic cells that are matured into an alpha-type-1-polarized phenotype (αDC1) using a specific cytokine cocktail (including IL-1β, TNF-α, IFN-α, IFN-γ, and poly-I:C). These matured cells are then loaded with apoptotic bodies from the allogeneic LNCaP prostate cancer cell line, which serves as a source for multiple prostate-specific antigens, including prostatic acid phosphatase (PAP) and prostate-specific membrane antigen (PSMA). Administered intradermally, the vaccine aims to stimulate a robust Th1-polarized cytotoxic T-lymphocyte response against the patient's tumor cells. It has been evaluated in clinical trials in combination with androgen ablation therapy.
