Overview

Allogeneic anti-CD19 CAR T cells are an "off-the-shelf" cellular immunotherapy engineered to express a chimeric antigen receptor (CAR) targeting the CD19 antigen, which is prevalent on B-cell malignancies. Unlike autologous CAR T therapies that utilize a patient's own cells, allogeneic products are derived from healthy donors, allowing for immediate availability and standardized manufacturing. A major challenge for these products is host-mediated allo-rejection, which limits their persistence. To address this, researchers are exploring engineering strategies such as the genomic silencing of HLA class I and II molecules (HLA-S CAR) to evade detection by the host immune system. Research presented at ASGCT 2026 focuses on developing quantitative assays to measure the survival and persistence of these engineered cells under alloreactive immune pressure.
