Overview

This experimental combination therapy regimen, developed by the National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK), is designed to induce donor-specific immunologic hyporesponsiveness in kidney transplant recipients. The protocol involves the administration of Thymoglobulin (a rabbit-derived anti-thymocyte globulin) for T-cell depletion, sirolimus (an mTOR inhibitor) for maintenance immunosuppression, and an infusion of allogeneic donor bone marrow cells. The primary objective of this approach is to promote graft tolerance and reduce the long-term reliance on standard immunosuppressive drugs, which are often associated with chronic nephrotoxicity and other systemic side effects.
