Overview

Allogeneic bone marrow-derived human mesenchymal stromal cells (MSCs) are multipotent stromal cells isolated from the bone marrow of healthy donors. These living cell therapies are capable of differentiating into various cell types including osteoblasts, chondrocytes, myocytes, and adipocytes. Their primary therapeutic mechanisms include immunomodulation (suppression or modulation of immune responses), anti-inflammatory effects, secretion of trophic factors that promote tissue repair and regeneration, antioxidative activity, antiapoptotic effects (preventing programmed cell death), and angiogenesis promotion. Allogeneic BM-MSCs have been investigated in a wide range of clinical indications such as steroid-refractory acute graft-versus-host disease (SR-aGVHD), autoimmune diseases (e.g., Crohn's disease, multiple sclerosis), degenerative disorders (e.g., degenerative disc disease), cardiovascular conditions (e.g., myocardial infarction), neurodegenerative diseases (e.g., Parkinson’s disease and Alzheimer’s disease), type 1 diabetes mellitus, retinal degenerative diseases, and more[1][3][4][5][8]. The first FDA-approved therapy using this modality is Ryoncil for pediatric SR-aGVHD[4][5][7][8].
