Overview

Allogeneic CCR5Δ32 stem cell transplantation is a specialized hematopoietic stem cell transplant (HSCT) procedure used to treat patients with both HIV-1 infection and hematologic malignancies. The procedure utilizes donor cells that are homozygous for the CCR5Δ32 mutation, a 32-base-pair deletion in the C-C chemokine receptor type 5 (CCR5) gene. This mutation results in the absence of functional CCR5 receptors on the surface of CD4+ T cells, which are the primary coreceptors for HIV-1 entry. By replacing the patient's immune system with these resistant cells, the procedure can lead to long-term HIV remission or cure, as demonstrated in cases like the Berlin, London, and Dusseldorf patients. It is typically performed when a patient requires a transplant for a life-threatening blood cancer, such as leukemia or lymphoma.
