Overview

Allogeneic CD123 CAR-NK cell therapy is a novel cellular immunotherapy designed for the treatment of relapsed/refractory acute myeloid leukemia (R/R AML). It utilizes natural killer (NK) cells, genetically engineered with a chimeric antigen receptor (CAR) targeting the CD123 antigen, which is highly and consistently expressed on AML blast cells and leukemia stem cells. This 'off-the-shelf' allogeneic approach aims to overcome challenges associated with CAR-T cell therapy in AML, such as target antigen heterogeneity and treatment-related toxicities, by leveraging NK cells' inherent tumor-killing mechanisms and favorable safety profile. The therapy involves generating CD123 CAR-NK cells via lentiviral transduction from peripheral blood mononuclear cells (PBMCs) of a single donor, followed by cryopreservation for clinical use. Patients receive a preconditioning regimen before multiple administrations of the CAR-NK cells.
