allogeneic CD4+ CD25+ regulatory T cells (gp120-activated), also known as ATreg, is an adoptive cell therapy developed by ActiTrexx GmbH. The product consists of donor-derived (allogeneic) regulatory T cells (Tregs) that are isolated and subsequently activated ex vivo using the HIV-1 envelope glycoprotein gp120. This activation process, mediated by gp120 binding to the CD4 receptor on Tregs, induces a stable and potent suppressive phenotype. The therapy is primarily being investigated for the prevention and treatment of acute graft-versus-host disease (aGvHD) in adult patients undergoing allogeneic hematopoietic stem cell transplantation (HSCT) for various hematologic malignancies, including acute myeloid leukemia and myelodysplastic syndrome. By administering these pre-activated Tregs early after transplantation, the therapy aims to suppress the donor-derived effector T cell response against host tissues while preserving the beneficial graft-versus-leukemia effect.
