Allogeneic cultured thymus tissue chips are a tissue-engineered therapeutic product developed for the treatment of congenital athymia, including complete DiGeorge syndrome. The therapy involves the transplantation of donor thymus tissue that has been processed and cultured for approximately 12 to 21 days. This culture period is critical as it depletes mature donor T-cells, thereby minimizing the risk of graft-versus-host disease (GvHD), while maintaining the viability of thymic epithelial cells and the thymic stroma. Once transplanted, typically into the quadriceps muscle, the tissue provides a functional microenvironment for the recruitment and maturation of host-derived T-cell progenitors, facilitating immune reconstitution in children born without a functional thymus.
