Overview

Allogeneic cytokine-induced killer (CIK) cells are an ex vivo expanded cell-based immunotherapy developed by Singapore General Hospital for the treatment of hematological malignancies relapsing after allogeneic hematopoietic stem cell transplantation (allo-HSCT). These cells are generated from donor peripheral blood mononuclear cells through sequential stimulation with interferon-gamma (IFN-γ), anti-CD3 monoclonal antibodies, and interleukin-2 (IL-2). This process results in a heterogeneous cell population dominated by CD3+CD56+ T cells that possess natural killer (NK)-like functional properties. Unlike conventional T cells, CIK cells exert potent antineoplastic activity in a major histocompatibility complex (MHC)-unrestricted manner, primarily mediated by the engagement of the natural killer group 2 member D (NKG2D) receptor with its ligands on malignant cells. This therapy aims to enhance the graft-versus-leukemia (GVL) effect while minimizing the risk of graft-versus-host disease (GVHD) compared to standard donor lymphocyte infusions (DLI).
