Allogeneic cytotoxic T lymphocytes (CTLs) are an investigational adoptive cell therapy developed by the University of Colorado, Denver, for the treatment of recurrent primary brain tumors. Unlike traditional tumor-infiltrating lymphocyte (TIL) therapy which typically uses a patient's own cells, this approach utilizes T cells derived from a donor (allogeneic). These cells are expanded and activated ex vivo using muromonab-CD3 (OKT3) and interleukin-2 (aldesleukin). The therapy is administered directly into the surgical cavity (intracavitary) following tumor resection, often in combination with additional aldesleukin to maintain T-cell activity and promote an anti-tumor immune response within the central nervous system environment. This strategy aims to bypass the immunosuppressive tumor microenvironment and provide a potent, targeted immune attack against residual cancer cells.
