Allogeneic dendritic cells (alloDCs) are a type of "off-the-shelf" cellular immunotherapy used primarily as an immunostimulatory adjuvant in cancer treatment. Unlike autologous dendritic cell vaccines, which are derived from the patient's own cells and loaded with specific antigens, alloDCs are derived from healthy donors and are typically administered intratumorally without being pre-loaded with tumor antigens. Their primary mechanism involves the secretion of potent pro-inflammatory cytokines (such as IL-12) and chemokines (such as CXCL10) upon injection. This creates an inflammatory milieu that recruits and activates the patient's endogenous dendritic cells, which then engulf tumor antigens in situ and migrate to lymph nodes to prime a systemic, tumor-specific T-cell response. This approach, notably developed by researchers at Uppsala University and commercialized as ilixadencel by Mendus (formerly Immunicum), aims to overcome the limitations of autologous therapies, such as high costs and manufacturing delays.
