The allogeneic dendritic cell cancer vaccine (DCCV) is an engineered cell therapy platform developed by Renovaro Biosciences for the treatment of various solid tumors. This platform utilizes dendritic cells derived from CD34+ hematopoietic stem cells (HSCs) that are genetically modified using a tri-cistronic lentiviral vector to overexpress CD93, CD40L (CD154), and CXCL13. The engineered dendritic cells are "educated" through co-culture with whole tumor lysates, allowing them to present a broad spectrum of tumor-associated antigens. The vaccine's mechanism of action involves the stimulation of both innate and adaptive immune responses, leading to the proliferation and tumor infiltration of CD4+ T cells, CD8+ T cells, and NK cells. Preclinical studies in humanized mouse models have demonstrated significant tumor regression in pancreatic cancer, and the platform is being explored for other indications including triple-negative breast cancer and hepatocellular carcinoma.
