Allogeneic EBV-specific DC-CTL (dendritic cell-activated cytotoxic T lymphocytes) is an adoptive cellular immunotherapy being developed for the treatment of Epstein-Barr virus (EBV)-associated lymphoproliferative diseases (EBV-LPD). The therapy involves generating EBV-specific cytotoxic T lymphocytes (CTLs) from healthy related donors (or HLA-haploidentical donors) by stimulating them with donor-derived dendritic cells (DCs) loaded with EBV-specific antigen peptides. These mature, antigen-primed CTLs are then expanded ex vivo and infused into the patient, where they specifically target and eliminate EBV-infected cells presenting viral latency proteins (such as LMP1, LMP2, and EBNA1) on their surface, thereby restoring anti-viral and anti-tumor immune surveillance without inducing significant graft-versus-host disease (GVHD).
