Allogeneic ex-vivo expanded adult limbal stem cells is an investigational cell therapy developed for the treatment of severe corneal ulcers, particularly trophic ulcers that are refractory to conventional treatments. This therapy involves the isolation of limbal stem cells from a healthy donor, which are then expanded ex-vivo, typically on a human amniotic membrane (hAM) carrier, to create a transplantable graft. The primary mechanism of action is the restoration of the limbal stem cell niche and the corneal epithelium. By providing a fresh source of progenitor cells, the transplant enables the repopulation of the ocular surface, differentiation into mature corneal epithelial cells, and the restoration of corneal transparency and barrier function. This allogeneic approach is especially critical for patients with bilateral limbal stem cell deficiency (LSCD), where autologous cell sources are unavailable. The development is led by the Fundacion Publica Para La Investigacion Biosanitaria De Andalucia Oriental Alejandro Otero (FPS) within the Andalusian Public Health System.
