Overview

The allogeneic GM-CSF-secreting breast cancer vaccine is an investigational cell-based immunotherapy developed at the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins. It consists of two irradiated allogeneic breast cancer cell lines (2T47D-V and 3SKBR3-7) that have been genetically modified to secrete human granulocyte-macrophage colony-stimulating factor (GM-CSF). The vaccine is designed to function as an immune primer; once injected intradermally, the secreted GM-CSF recruits and activates dendritic cells at the injection site. These dendritic cells then ingest the irradiated tumor cells, process their associated antigens (including HER2/neu), and present them to T cells to stimulate a systemic, tumor-specific immune response. Clinical development has focused on using this vaccine in combination with immunomodulatory agents like cyclophosphamide and targeted therapies like trastuzumab to treat HER2-overexpressing breast cancer.
