This therapeutic intervention refers to allogeneic hematopoietic stem cell transplantation (HSCT) using unrelated or partially matched donors, as investigated by the Masonic Cancer Center at the University of Minnesota. The protocol is designed for patients with previously treated lymphoma, multiple myeloma, or chronic lymphocytic leukemia. The procedure involves a conditioning regimen of chemotherapy (busulfan and cyclophosphamide) and total body irradiation to eradicate malignant cells and prevent graft rejection. Following conditioning, donor-derived stem cells (from bone marrow or umbilical cord blood) are infused to reconstitute the patient's hematopoietic system and induce a graft-versus-tumor (GVT) effect, where donor immune cells target residual cancer. Post-transplant immunosuppression with agents like methotrexate and cyclosporine is typically employed to mitigate graft-versus-host disease (GVHD).
