Allogeneic hematopoietic stem cell transplantation (HSCT) is a cellular therapy used to treat high-risk or relapsed/refractory hematologic malignancies. In the context of the Hannover Medical School trial (NCT00606723), it involves the infusion of hematopoietic stem cells derived from bone marrow or peripheral blood of matched sibling, matched unrelated, or haploidentical donors. The therapeutic effect is twofold: first, the donor cells reconstitute the patient's hematopoietic and immune systems following intensive conditioning (such as the FLAMSA regimen); second, the donor-derived T-cells and NK cells exert a "graft-versus-leukemia" (GvL) effect, targeting and destroying residual leukemic cells. This trial specifically focuses on optimizing donor selection and conditioning strategies for children and young adults with acute myeloid leukemia (AML).
