Allogeneic mesothelin-targeting CAR-NKT cells (AlloMCAR-NKT) are an off-the-shelf cellular immunotherapy developed using a hematopoietic stem and progenitor cell (HSPC) engineering platform. These cells are engineered to express a chimeric antigen receptor (CAR) specific for mesothelin, a protein overexpressed in various solid tumors including endometrial carcinoma. Unlike conventional CAR-T cells, these CAR-NKT cells utilize the innate-like properties of invariant natural killer T (iNKT) cells, allowing them to target both mesothelin-positive tumor cells and CD1d-expressing immunosuppressive cells within the tumor microenvironment, such as tumor-associated macrophages (TAMs) and myeloid-derived suppressor cells (MDSCs). The allogeneic nature and HSPC-based production method aim to provide a scalable, safe, and effective treatment for aggressive, chemoresistant solid tumors with a reduced risk of graft-versus-host disease (GvHD).
