An allogeneic myeloma vaccine is a type of cancer immunotherapy designed to stimulate the immune system to recognize and attack multiple myeloma cells. The "allogeneic" designation means the vaccine is derived from donor (not patient) cells, typically using irradiated or genetically modified myeloma cell lines as the antigen source. These vaccines are often engineered to express immune-stimulating factors such as granulocyte-macrophage colony-stimulating factor (GM-CSF), which enhances antigen presentation and T-cell activation. The primary indication is for multiple myeloma, either in combination with other agents like lenalidomide or as maintenance therapy following autologous stem cell transplantation. The goal is to induce anti-myeloma immunity, reduce minimal residual disease, and prolong remission.
