This investigational cell-based immunotherapy is a combination treatment developed by the Centre Hospitalier Universitaire de Besançon for the treatment of EGFR-positive metastatic gastrointestinal cancers with liver involvement. The regimen consists of allogeneic (donor-derived) haploidentical natural killer (NK) cells administered via hepatic intraarterial infusion, combined with the anti-EGFR monoclonal antibody cetuximab. The therapeutic strategy relies on the synergistic effect of direct NK cell-mediated cytotoxicity and cetuximab-induced antibody-dependent cellular cytotoxicity (ADCC). To support the persistence and activity of the infused NK cells, the protocol includes a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine, followed by the administration of high-dose interleukin-2 (IL-2).
