CTM-N2D is an investigational, allogeneic cell therapy product consisting of γδ T cells engineered to express a chimeric antigen receptor (CAR) that targets NKG2D ligands (NKG2DL). These ligands are stress-induced proteins commonly expressed on the surface of many tumor types but not on normal tissues. The γδ T cells are derived from healthy donors and expanded ex vivo before being modified via mRNA electroporation to express the CAR construct. Upon administration, CTM-N2D is designed to recognize and bind tumor cells expressing NKG2DL, leading to targeted cytotoxicity against these cancerous cells. This approach leverages both the innate anti-tumor properties of γδ T cells and the specificity conferred by the engineered CAR targeting NKG2DL. The primary developer is CytoMed Therapeutics, which is conducting phase I clinical trials in patients with advanced solid tumors or hematological malignancies[3][4][7].
