Allogeneic PCART7 is an "off-the-shelf" chimeric antigen receptor (CAR) T-cell therapy being developed by MediSix Therapeutics for the treatment of CD7-positive T-cell malignancies, including T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LL). The therapy targets CD7, a surface antigen highly expressed on malignant T-cells. To enable the production of anti-CD7 CAR-T cells and their use in an allogeneic setting, MediSix utilizes its proprietary Protein Expression Blocker (PEBL) technology. This technology downregulates surface expression of CD7 on the CAR-T cells themselves to prevent fratricide (self-destruction) and suppresses the TCR/CD3 complex to reduce the risk of graft-versus-host disease (GvHD). The therapy is designed to induce potent cytotoxicity against CD7-positive malignant cells and is currently in the IND-enabling stage of development.
