Overview

Allogeneic peripheral blood-derived CD56+ natural killer (NK) cells are an adoptive cell therapy product consisting of NK cells isolated from healthy donor peripheral blood. These cells are expanded and activated ex vivo to enhance their anti-tumor potency. In clinical protocols, such as those led by Hospital Universitario La Paz, the cells are co-cultured with K562-mb15-41BBL feeder cells, which express membrane-bound interleukin-15 (IL-15) and 4-1BB ligand (4-1BBL) to stimulate proliferation and cytotoxic activity. Once infused, these activated NK cells exert direct anti-tumor effects through the release of cytotoxic granules (perforin and granzymes) and the secretion of pro-inflammatory cytokines, targeting malignant cells that often downregulate MHC class I molecules to evade T-cell detection. This therapy is being investigated for the treatment of sarcomas in pediatric and young adult populations.
