Overview

This therapeutic approach, developed and investigated at the M.D. Anderson Cancer Center, involves the use of allogeneic peripheral blood progenitor cells (PBPCs) for the treatment of recurrent or metastatic breast cancer. The procedure utilizes a reduced-intensity ("mini") conditioning regimen, typically consisting of fludarabine and melphalan, to suppress the recipient's immune system sufficiently to allow for donor cell engraftment without the high toxicity associated with traditional myeloablative conditioning. The primary therapeutic mechanism is the induction of a graft-versus-tumor (GVT) effect, where donor-derived immune cells (such as T cells and NK cells) recognize and eliminate residual malignant breast cancer cells in the recipient. This strategy aims to provide a long-term immunotherapy-based remission for patients with advanced disease.
