The allogeneic prostate cell line-loaded DC1 vaccine is an experimental autologous dendritic cell-based immunotherapy developed by the Roswell Park Comprehensive Cancer Center. The vaccine utilizes patient-derived dendritic cells that are polarized in vitro to an alpha-type-1 (DC1) phenotype, which is specifically designed to produce high levels of interleukin-12 (IL-12) to favor the induction of potent Th1 and cytotoxic T-lymphocyte (CTL) responses. These DC1 cells are loaded with a broad spectrum of tumor-associated antigens derived from the allogeneic LNCaP prostate cancer cell line, which has been induced to undergo apoptosis. By presenting multiple antigens simultaneously, the vaccine aims to overcome tumor escape mechanisms and stimulate a robust immune response against the patient's prostate cancer. It has been primarily investigated for patients with recurrent prostate cancer who exhibit rising PSA levels after local therapy failure, often administered in conjunction with androgen ablation therapy.
