Allogeneic prostate vaccines are therapeutic cancer vaccines composed of whole cells derived from multiple allogeneic (non-self) human prostate cancer cell lines. These cells are typically irradiated to prevent proliferation and may be genetically modified to secrete immunostimulatory factors such as granulocyte-macrophage colony-stimulating factor (GM-CSF). The goal is to stimulate the patient's immune system—particularly T cell responses—against a broad array of tumor-associated antigens present in the donor cell lines. Notable examples include Onyvax-P, which uses three irradiated allogeneic prostate cancer cell lines and is administered with bacille Calmette-Guérin as an adjuvant for initial doses[1][2][5]. Another example is GVAX-PCa, which consists of two genetically modified allogeneic cell lines secreting GM-CSF[1][3]. These vaccines have been investigated primarily for hormone-resistant or castration-resistant prostate cancer. Clinical studies have shown favorable safety profiles and some evidence of delayed disease progression or improved survival compared to historical controls[1][2][4].
