allogenic T cell-depleted hematopoietic stem cell transplantation is a cell therapy protocol developed by Leland Metheny for the treatment of engraftment failure following an initial hematopoietic stem cell transplant. The procedure involves the infusion of a booster dose of donor hematopoietic stem cells that have been ex vivo processed to enrich for CD34+ progenitor cells and deplete CD3+ T cells, typically using the CliniMACS Reagent System. This selective depletion is intended to promote hematopoietic recovery and donor chimerism while minimizing the risk of graft-versus-host disease (GvHD), a common and serious complication of allogeneic transplantation caused by donor T-cell reactivity against recipient tissues.
