Overview

Allonix Therapeutics is developing a first-in-class oral small molecule agonist of liver receptor homologue-1 (LRH-1), a nuclear hormone receptor also known as nuclear receptor subfamily 5 group A member 2 (NR5A2). LRH-1 plays a critical role in regulating inflammation, cholesterol homeostasis, and cellular metabolism. The drug is designed to bind to LRH-1 in intestinal cells to stimulate the local production of corticosteroids, thereby reducing inflammation and promoting the healing and regeneration of damaged gut tissue. This localized approach is intended to treat inflammatory bowel disease (IBD), specifically ulcerative colitis, while minimizing the systemic side effects associated with traditional corticosteroid therapies. The program is currently in the preclinical stage and has received support from the Crohn's & Colitis Foundation's IBD Ventures program.
