AlloNK® + obinutuzumab is an investigational combination therapy developed by Artiva Biotherapeutics for the treatment of autoimmune diseases, including systemic lupus erythematosus (SLE) and lupus nephritis (LN). The combination consists of AlloNK (AB-101), a non-genetically modified, allogeneic, cryopreserved, off-the-shelf natural killer (NK) cell therapy, and obinutuzumab, a type II anti-CD20 monoclonal antibody. The mechanism of action involves AlloNK cells enhancing the antibody-dependent cellular cytotoxicity (ADCC) of obinutuzumab to drive deep depletion of CD20-expressing B cells. This approach aims to provide a potent B-cell targeted therapy without the need for complex genetic engineering or autologous manufacturing. The program is currently being evaluated in a Phase 1 clinical trial (NCT06265220) in patients with active lupus nephritis.
