Overview

Alloreactive cytotoxic T lymphocytes (alloCTL) combined with interleukin-2 (IL-2) is an experimental cellular immunotherapy developed by the Jonsson Comprehensive Cancer Center (UCLA) for the treatment of recurrent malignant gliomas and malignant meningiomas. This biologic therapy consists of ex vivo activated alloreactive cytotoxic T lymphocytes that are sensitized to the patient's major histocompatibility complex (MHC) proteins. The therapy involves generating alloCTL from MHC-disparate donor lymphocytes through mixed lymphocyte reactions over a 2-3 week period. These sensitized cells are then administered intratumorally or into the tumor cavity along with IL-2, which serves as a growth factor to support the survival and anti-tumor activity of the T cells. The mechanism of action relies on the selective recognition and killing of glioma cells expressing the targeted MHC proteins by the donor-derived cytotoxic T cells.
