Allovectin-7 is a gene therapy agent developed for the treatment of cancer, primarily malignant melanoma. It consists of a plasmid/lipid complex containing DNA sequences encoding HLA-B7 and β2-microglobulin, which are components of the major histocompatibility complex (MHC) class I. The drug is administered intratumorally and aims to increase MHC class I expression on tumor cells, thereby enhancing recognition and destruction by the immune system. Its mechanism involves inducing both cytotoxic T-cell and innate immune responses against tumor cells through increased antigen presentation and localized immune activation. Developed by Vical, Allovectin-7 reached phase 3 clinical trials for metastatic melanoma but was discontinued after failing to demonstrate improved objective response rate or overall survival compared to chemotherapy. The mechanism is not specific to melanoma, suggesting potential applicability in other solid tumors[1][2][6][8].
