Overview

ALLR3 (also known as UK ALLR3) is a multi-drug chemotherapy reinduction regimen specifically designed for the treatment of children with relapsed or refractory B-cell precursor acute lymphoblastic leukemia (ALL). The regimen is a combination of four primary agents: vincristine (a microtubule inhibitor), mitoxantrone (a DNA intercalator and topoisomerase II inhibitor), dexamethasone (a corticosteroid), and asparaginase (an enzyme that depletes asparagine). Originally developed by the Children's Cancer and Leukaemia Group (CCLG) in the United Kingdom, ALLR3 has been adopted globally by pediatric oncology consortia, including the Children's Oncology Group (COG), as a standard-of-care backbone. In contemporary clinical research, it frequently serves as the active comparator for evaluating novel targeted therapies, such as Pfizer's Phase 2 study of the antibody-drug conjugate inotuzumab ozogamicin.
