Overview

ALMB-0166 is a humanized monoclonal antibody that targets and inhibits connexin 43 (Cx43) hemichannels, specifically blocking their activity in astrocytes. By closing these hemichannels, ALMB-0166 prevents the release of pro-inflammatory molecules following acute spinal cord injury, thereby reducing local inflammation and scar formation. This mechanism supports neuronal regeneration and functional recovery after spinal cord injury. The drug is being developed primarily for the treatment of acute spinal cord injuries and has received orphan drug designation from the US FDA for this indication. It is currently in phase I clinical trials for spinal cord injuries, with preclinical development ongoing for osteoarthritis and early research in stroke[1][2][3][4][5][7].
