ALMB-0168 is a first-in-class, humanized monoclonal antibody that acts as an agonist of the connexin 43 (Cx43; also known as gap junction alpha-1 protein) hemichannel. It is being developed for the treatment of osteosarcoma, particularly in patients who have progressed after standard chemotherapy. The drug works by activating Cx43 hemichannels on cell membranes, which leads to the release of tumor-inhibiting cytokines and suppression of tumor growth and metastasis in preclinical models. ALMB-0168 has demonstrated efficacy in inhibiting both osteosarcoma and breast cancer bone metastases in preclinical studies. It has received rare pediatric disease designation and orphan drug designation from the U.S. FDA for osteosarcoma[1][2][3][4][7].
