almorexant

Almorexant is a first-in-class, orally active small molecule that acts as a dual orexin receptor antagonist, competitively inhibiting both orexin receptor type 1 (OX1R) and orexin receptor type 2 (OX2R) in the brain. By blocking these receptors, which are involved in the regulation of wakefulness and arousal, almorexant was developed to promote sleep and treat primary insomnia. The drug was originally discovered by Actelion and later co-developed with GlaxoSmithKline for sleep disorders such as insomnia. Almorexant reached Phase III clinical trials but its development was discontinued in 2011 due to concerns over its side effect profile[2][4][5].
